The functioning of cells depends on the ability of communication networks of cell societies to transmit essential information as signaling pathways, and these cellular responses are mediated by numerous biomolecular interactions. Including cancer, chronic inflammatory syndromes and diabetes, all result from the malfunction of one or more steps in the signaling pathway. Therefore, the atomic-resolution exploration of protein-protein interactions is critical for developing effective strategies for therapeutic interventions against human diseases.

We introduce nuclear magnetic resonance spectroscopy (NMR), the most common technique for studying protein-protein interactions. NMR provides atomic resolution information about binding interfaces, intermolecular affinity, and binding-induced conformational changes, etc. Notably, continued advancements in solution and solid-state NMR techniques have established this method as a fundamental research tool to accelerate our biochemical research of signal transduction pathways.
