α-Ketoglutarate dehydrogenase (α-KGDH, EC 1.2.4.2) is one of the key regulatory enzymes of the citric acid cycle and is widely present in animals, plants, microorganisms, and cultured cells. It can catalyze the conversion of α-ketoglutarate, NAD+, and coenzyme A into succinyl coenzyme A, carbon dioxide, and NADH. The enzyme activity is determined by measuring the increase rate of NADH at 340 nm.
