Phospholipids are hydrolyzed by phospholipase D to form choline and phosphatidic acid, which is then oxidized by choline oxidase to form hydrogen peroxide. Hydrogen peroxide reacts with 4-aminophenazone and DAOS to form a blue dye. This dye has the maximum absorption peak at 600nm, and the absorption intensity is proportional to the phospholipid content in the sample.
