Under acidic conditions, copper is dissociated from ceruloplasmin and albumin. Ascorbic acid (reduced form) reduces the dissociated copper(II) ions to copper(I) ions, which then form a blue complex with the chromogen 3,5-DiBr-PAESA. The absorbance of the blue copper complex at 600nm is measured to calculate the concentration of serum copper.
