Total cholesterol (TC) includes free cholesterol and cholesterol esters. It refers to the sum of cholesterol contained in all lipoproteins in tissues. Esterase catalyzes the hydrolysis of cholesterol esters to produce free cholesterol (FC), which is then oxidized by cholesterol oxidase to generate 4-cholestenone and H2O2. Subsequently, it reacts with 4-aminoantipyrine to form a red quinone compound, which has a characteristic absorption peak at 510nm. By measuring the absorbance at 510nm, the TC content can be determined.
