3-Phosphoglycerate (G3P) is oxidized by 3-phosphoglycerate oxidase (GPO) to produce hydrogen peroxide (H2O2), which reacts with 4-aminoantipyrine to form a red quinone compound with a characteristic absorption peak at 510nm. By detecting the change in absorbance at 510nm, the activity of 3-phosphoglycerate oxidase (GPO) can be determined.
