Albumin carries a positive charge in a buffer solution at pH 4.2 and can combine with the negatively charged dye bromocresol green to form a blue-green complex in the presence of a non-ionic surfactant. The complex has an absorption peak at 630nm, and the color intensity is directly proportional to the concentration of albumin.
