Cholesterol Esterase (CHER) and Cholesterol Oxidase (CHOD) are chemically modified to reduce their reactivity with LDL, VLDL, and chylomicrons, selectively acting only with HDL-cholesterol. In the first step, LDL, VLDL, and chylomicrons are combined with sodium gluconate and sulfonated cyclodextrin complex. In the second step, chemically modified CHER and CHOD are used to directly measure HDL-cholesterol without the need to separate other lipoproteins. The modified CHER catalyzes the hydrolysis of cholesterol esters to form free cholesterol (FC), which is then oxidized by CHOD to form 4-cholestenone and H2O2. This reacts with 4-aminoantipyrine to form a red quinone compound with a characteristic absorption peak at 546nm, from which HDL-C content is determined.
