4-hydroxyproline (Hyp) is a non-essential amino acid and a major component of collagen tissue, found only in collagen and elastin in mammals but in many proteins in plants. Many diseases in animals are accompanied by changes in collagen metabolism, leading to changes in the content of hydroxyproline in blood, urine, and tissues. This kit uses acid hydrolysis to produce free HYP, which is further oxidized by chloramine T, and the oxidation product reacts with p-dimethylaminobenzaldehyde to show a purple-red color. By measuring the absorbance of the colored substance at 560nm, the HYP content can be determined.
