Potassium is detected through the enzymatic reaction catalyzed by potassium-dependent pyruvate kinase with the substrate phosphoenolpyruvate (PEP). The product pyruvate salt reacts with NADH to form coenzyme I (NAD+) under the action of lactate dehydrogenase (LDH), and the decrease in absorbance at 340nm is proportional to the potassium concentration.
