Lyophilized from sterile 100mM Glycine, 10mM NaCl, 50mM Tris, pH 7.5, 5 % trehalose and 5 % mannitol.
Bio-activity
Measured by its binding ability in a functional ELISA. Immobilized recombinant human CD112 at 20 μg/ml (100 μl/well) can bind biotinylated DNAM1 with a linear range of 0.078-5 μg/ml.
Molecular Mass
RecombinantHuman CD112/Fc chimera is a disulfide-linked homodimeric protein. The reduced monomer consists of 567 amino acids and has a calculated molecular mass of 62.3 kDa. As a result of glycosylation, the rh CD112/Fc monomer migRates as an approximately 70-80 kDa protein in SDS-PAGE under reducing conditions.
Endotoxin
< 1.0 EU per 1 microgram of protein (determined by LAL method).
Purity
> 95 % by SDS-PAGE.
Storage
In lyophilized state for 1 year (4°C); After reconstitution under sterile conditions for 3 months (-70°C). Avoid repeated freeze/thaw cycles.
Reconstitution
Reconstitute in sterile distilled water to a concentration of 0.1-1.0 mg/mL.

Background
Cytokine that binds to CD40/TNFRSF5 (PubMed:1280226). Costimulates T-cell proliferation and cytokine production. Its cross-linking on T-cells generates a costimulatory signal which enhances the production of IL4 and IL10 in conjunction with the TCR/CD3 ligation and CD28 costimulation (PubMed:8617933). Induces the activation of NF-kappa-B and kinases MAPK8 and PAK2 in T-cells. Induces tyrosine phosphorylation of isoform 3 of CD28 (PubMed:15067037). Mediates B-cell proliferation in the absence of co-stimulus as well as IgE production in the presence of IL4. Involved in immunoglobulin class switching (By similarity). Release of soluble CD40L from platelets is partially regulated by GP IIb/IIIa, actin polymerization, and an matrix metalloproteinases (MMP) inhibitor-sensitive pathway.
