Sodium is detected through the enzymatic reaction catalyzed by sodium-dependent β-galactosidase with the substrate ONPG (O-nitrophenyl-β-D-galactopyranoside). The product O-nitrophenol has an absorbance at 405nm that is proportional to the sodium concentration.
