ABTS is oxidized to green ABTS+ by an appropriate oxidizing agent, and the production of ABTS+ is inhibited in the presence of antioxidants. The total antioxidant capacity of the sample can be measured and calculated by determining the absorbance of ABTS+ at 734nm or 414nm. Trolox, a vitamin E analog, has antioxidant capacity similar to vitamin E and is used as a reference for the total antioxidant capacity of other antioxidants.
Additional Materials and Equipments Required
Microplate reader, 96-well plate, low-temperature centrifuge, water bath, pipette, mortar, anhydrous ethanol, and distilled water.
