Total Ascorbic Acid (TAA) includes both reduced and dehydroascorbic acid. Dehydroascorbic acid is reduced to reduced ascorbic acid, which then reduces ferric ions to ferrous ions. Ferrous ions react with 4,7-Diphenyl-1,10-phenanthroline to form a red complex with a characteristic absorption peak at 534nm. The color intensity is proportional to the content of reduced ascorbic acid, from which the total ascorbic acid content is calculated.

Microplate reader, 96-well plate, low-temperature centrifuge, water bath, pipette, mortar, methanol, anhydrous ethanol, and distilled water.
