Total Ascorbic Acid (TAA) includes both reduced and dehydroascorbic acid. Reduced ascorbic acid is easily oxidized to dehydroascorbic acid, which reacts with 2,4-dinitrophenylhydrazine to form a red complex soluble in sulfuric acid. This red substance has a maximum absorption peak at 520nm, from which the total ascorbic acid (TAA) content is calculated.

Plate reader, 96-well plate, concentrated sulfuric acid, mortar and pestle, ice, low-temperature centrifuge, pipette, and distilled water.
