Sugar substances are one of the important components of plant bodies and are also the main raw materials and storage substances for metabolism. Total sugars, also known as carbohydrates, include soluble monosaccharides, disaccharides, and insoluble starch, cellulose, chitin, etc. Total sugars are hydrolyzed into reducing sugars, which react with DNS reagent under alkaline conditions to form an amino compound that appears orange-red in an excess NaOH alkaline solution. After scanning from 500nm to 540nm, the maximum absorption peak is at 500nm, and within a certain concentration range, the content of reducing sugars is linearly related to the absorbance at 500nm. Based on the standard curve, the content of reducing sugars in the sample, i.e., the total sugar content, can be determined.
